you can only add 1 copy of non-haven sites to your location deck, so i'm affraid yes, it is unique

Is something that would prevent my opponent from playing its own copy of Ancient Deep-hold while my copy is in play?

no, but i guess Darksatin's question was related to having more than 1 ancient deep hold in play by the same player.

I know, technically, it is not unique because 2 players can have it in play at the same time as well as any other site. But in terms of deckbuilding, you can't add more than 1 copy of each non-haven site to location deck (deep mines is the only exception as far as i can remember)
